FACS off! Autonomous & high throughput single cell cloning

B-cell cloning is an industry standard method for reliable and reproducible production of monoclonal antibodies (mAb). A typical journey in mAb development involves B cells isolation prior to screening. For years, FACS has been the method of choice for semi-automated seeding of single cells into microwell plates. However, this method suffers from low throughput due to inherent difficulties to automate start up, calibration, sample/target exchange processes. Furthermore, FACS is labour-intensive as it requires continuous presence of skilled operator to monitor and troubleshoot the process. Hence, the need of an alternative approach that addresses the aforementioned shortcomings that are: throughput, clonability, and high operating costs.

In this webinar, we will present how cellenONE HT, an image-based single cell isolation platform, is fully automated using Green Button Go Scheduler so that it operates continuously without the need for an operator, effectively enabling 24/7 unattended single cell plating from one or multiple samples.
